(A) pAd5-CMV-rtTA-TRE-PminCMV contains two expression cassettes with rtTA and the transgene controlled by two different promoters, pCMV and TRE-PminCMV, respectively, in the same direction. (B) pAd5-CMV-rtTA2S-M2-TRE-PminCMV is similar to pAd5-CMV-rtTA-TRE-PminCMV except that rtTA was replaced by rtTA2S-M2. (C) pAd5-Bi-Tet-On contains two promoters placed on both sides of TRE, forming an autoregulatory loop. (D) pAd5-KRAB-Bi-Tet-On contains an additional TetR-KRAB expression cassette compared to pAd5-Bi-Tet-On.
