A 6 week, repeat intramuscular injection, regimen of PEG-PEI-ESO polyplexes produced widespread manifestation of dystrophin-positive fibers. Dystrophin immunolabeling of whole transverse sections of TA muscles of mdx mice is shown after 10 twice-weekly intramuscular injections of either 1 μg (A-B) or 5 μg (C-D) of ESO complexed with the NG-PEI2K(PEG550)10 copolymer. Muscles were harvested at 6 wks after the first of the 10 injections. High magnification views of four individual regions of each muscle section (boxed) are shown.
