Blockade of mitochondrial permeability transition (MPT). Vero-B4 cells were incubated in RPMI 1640 or a modified KH buffer containing glucose, a low Ca2+ concentration and a high inorganic phosphate concentration (KHG(Ca-,P+)) at 4°C for 168 hours and then rewarmed in RPMI 1640 at 37°C for 3 hours. Part of the cells were cold incubated in the presence of the iron chelator deferoxamine (1 mM) or of inhibitors of the mitochondrial permeability transition, trifluoperazine (tfp, 20 μM) and fructose (10 mM). A solvent control with ethanol was included (+ EtOH). Cell injury was assessed by release of lactate dehydrogenase directly after cold storage (168 h) and hourly during 3 h of rewarming (LDH; n = 4; ** p < 0.01 vs. RPMI 1640 at 171 h).
