Lack of impact of scFv on proliferation of HepG2 cells. A. Validation of MTT assay as measurement of growth of HepG2. HepG2 and HepG2.sh57 cell lines were grown for 4 days in culture. Manual counting with hematocytometer of trypsinized cells correlated strongly with MTT OD450 (R2 = 0.99). B. Effect of scFv on cell line proliferation. MTT OD450 for HepG2 cultured for 2 or 4 days in the presence or absence of 2E10, 3E11, 3D8, 4G5, and 2G9 at 1 μg/ml showed no evidence of growth inhibition.
