Affinity purification of sCD4. a SDS-PAGE analysis of purified sCD4. 7 × 106 gene-modified 293 T cells were seeded into five 10 cm dishes. Media were exchanged to fresh media 1 day after plating. On the second day, media were replaced with 10 ml fresh media containing 1 % FBS and the cells were incubated for 2 additional days before sCD4 was purified from the supernatants. Samples from unmodified cells (mock), gene-modified cells (LV-sCD4) and purified sCD4 were analyzed by SDS-PAGE followed by Coomassie blue staining. b Comparison of purified sCD4 to an sCD4 standard from the National Institutes of Health. Equal volumes of the standard and purified sCD4 were analyzed by SDS-PAGE followed by Coomassie blue staining. Results are representative of 3 independent experiments
