Thermal inactivation profiles (a) and half-lives (t1/2; b) of the wild-type and variants of DHAP. The enzyme (0.5 μg) was diluted in borate/NaOH buffer (pH 9.6) and incubated at 60 °C for various times. Then, the survival activity was assayed in a 400 μL reaction mixture (borate/NaOH buffer, pH 9.6, 2 mM CaCl2) containing 1% casein as substrate for 10 min. The relative activity was normalized to the initial activity (100%). The data are expressed as the average value from three independent experiments
