Comparison of actual reaction fluorescence readings to those predicted by LRE modeling. Although the DyNAmo series was selected for presentation, all three series presented in Figure 2 produced similar results. The average F0 calculated from the LRE window (designated by red circles, see Figure 3) was used to calculate the predicted FC across the entire amplification profile using equation 7 (circles) and plotted with the actual FC readings (dots). The numerical summaries presented as inlays illustrate the high degree of precision that can be achieved, with the average percent difference of < 0.5% for FC readings within the LRE window. FC: actual reaction fluorescence, pFC: predicted reaction fluorescence, Diff: difference between actual and predicted FC, %pFC: difference expressed as a percentage of the predicted FC.
