Activation of STAT3 by E. coli derived viral IL-10. STAT3 phosphorylation (STAT3-pY705) was analyzed by immunoblot of protein extracts from human Daudi cells (HCMV IL-10; A) and J774.1 mouse macrophages (EBV IL-10; B) treated with different concentrations of bacteria-derived viral IL-10. Total STAT3 was used to ensure equal protein loading in all lanes (double bands in Daudi cells represent STAT3 isoforms α and β). Periplasmic and cytoplasmic fractions of pUC19 transformed E. coli BL21 (DE3) cells and commercial viral IL-10 proteins served as controls. One experiment representative of two is shown.
