  1. Total primary clones were calculated from standard serial dilutions. For each library and vector, ~50 colonies were randomly chosen for plasmid isolation. Plasmids were digested and run on agarose gels to calculate respective insert sizes. Clone size differences between libraries are not considered significant (unpaired, two tailed t tests between pDONR222 and pDEST22 libraries: hormone library - P value = 0.7427, t = 0.3293 with 94 degrees of freedom; photoperiod library P value = 0.6213, t = 0.4955 with 104 degrees of freedom).
