High-density transfection of Hek293 cells. Cells were re-suspended in 20 mL of fresh HyQSFM4TransFx293 medium to a final density of 5 × 106 cells/mL and transfected with 2 μg DNA and 4 μg PEI in 125 mL Erlenmeyer flask at 34°C and 37°C. Transfections were done at 0, 48, and 96 hours post-inoculation. a: total cell density and viability; b: rFVIII activity measured by chromogenic assay; and c: rFVIII concentration measured by ELISA.
